2、 诊治及预测临床预后中的价值。方法:回顾性分析 2014 年 1 月至 2022 年 1 月于东部战区总医院泌尿外科就诊的 310 例男性患者。其中健康对照组 80 例,BPH 组 80 例,PCa 组 150 例(均接受根治性前列腺切除术,未接受化疗、放疗或雄激素剥夺治疗)。留取受试者的血液标本,采用实时荧光定量 PC 检测 310 名受试者的血清 mi-129 表达水平并分析其有无统计学差异。收集 150 例 PCa 患者的原发性 PCa 组织和对应的癌旁前列腺组织(距离病灶3 cm)样本,所有 PCa 组织的病理学分级均由经验丰富的病理学家完成。记录每位患者的人口学数据和临床参数,包括年
3、龄、体质指数(BMI)、临床分期、Gleason 评分、手术边缘状态、血管淋巴浸润状态、精囊浸润状态和生化复发(BC)情况。采用 T-qPC 测定组织样本中的 mi-129 含量,分析其与上述人口学数据和临床参数的关系。选用 PC-3 和 DU-145 细胞系并分别转染 mi-129 precursor 或 mi-129 scramble。通过细胞增殖测定和蛋白质印迹分析评估各组细胞系的增殖能力及相关蛋白的表达水平,比较分析其结果。结果:与健康男性和 BPH 患者相比,PCa 患者的血清 mi-129 表达水平显著降低(P 0 01);与癌旁前列腺组织相比,PCa 组织中 mi-129 表达水
4、平显著降低(P 均 0 01)。mi-129 的表达水平与术前血清特异性抗原(PSA)水平(P 0 001)、组织学分级(P 0 001)、病理分期(P 0 001)、Gleason 评分(P 0 001)、淋巴结转移(P=0 02)、血管淋巴侵袭(P=0 021)和生化复发(P=0 001)均呈负相关。Kaplan-Meier 分析表明,PCa 组织中低 mi-129 表达水平与较低的无 BC 生存率密切相关。多变量生存分析表明,mi-129 表达水平可能是 PCa 患者无 BC 生存率的独立预后指标(P 0 001)。高水平的mi-129 可能通过调控细胞周期调节蛋白的表达显著抑制 PCa
