2、州646000;2.西南医科大学口颌面修复重建和再生泸州市重点实验室,西南医科大学口腔医学研究所,四川泸州646000)摘要目的:探讨饰胶蛋白聚糖基因(decorin,DCN)过表达对口腔鳞癌(oral squamous cell carcinoma,OSCC)裸鼠荷瘤中表皮生长因子受体(epidermal growth factor receptor,EGFR)、骨髓细胞瘤病毒癌基因(cellular-myelocytomatosis viraloncogene,C-Myc)和细胞周期蛋白依赖性激酶抑制剂(cyclin dependent kinase inhibitor,p21)表达水平的
3、影响。方法:通过脂质体转染法上调人口腔鳞癌细胞(HSC-3)中DCN基因的表达,使用裸鼠作为OSCC疾病模型构建载体,建立OSCC裸鼠荷瘤模型。H-E法确定各组裸鼠荷瘤组织的病理分级,免疫组织化学染色检测DCN过表达后各组裸鼠荷瘤组织中EGFR、C-Myc和p21蛋白的表达差异,RT-qPCR及Western印迹定量检测DCN过表达后各组裸鼠荷瘤组织中EGFR、C-Myc和p21在转录水平和蛋白水平上的表达差异,明确DCN过表达对OSCC裸鼠荷瘤组织中EGFR、C-Myc和p21表达的影响。采用SPSS 20.0软件包对数据进行统计学分析。结果:H-E染色结果显示,OSCC动物疾病模型构建成功
4、。重量分析结果显示,质粒组裸鼠荷瘤组织轻于空载组和未转染组(P0.05)。IHC结果显示,DCN、EGFR、C-Myc和p21蛋白在各组裸鼠荷瘤组织中均有表达,质粒组中DCN、EGFR和C-Myc蛋白的表达水平差异有统计学意义(P0.05)。RT-qPCR和Western印迹结果显示,DCN、EGFR、C-Myc和p21在各组裸鼠荷瘤组织中均有不同程度表达(P0.05)。结论:上调DCN可抑制OSCC裸鼠荷瘤的生长。在OSCC裸鼠荷瘤组织中,DCN过表达下调EGFR及C-Myc的表达,上调p21的表达。DCN可能通过以上机制调节OSCC的发生、发展,发挥抑癌作用。关键词饰胶蛋白聚糖;口腔鳞癌;

11、tissues of nude mice in the plasmid group were significantly lighter than those inthe empty vector group and non-transfected group(P0.05).IHC results showed that DCN,EGFR,C-Myc and p21 proteinswere expressed in the tumor-bearing tissues of nude mice in each group,the expression of DCN,EGFR and C-Myc
12、 pro-teins in the plasmid group was significantly different from the other groups(P0.05).RT-qPCR and Western blot results showed that DCN,EGFR,C-Myc andp21 were expressed in diffrent degrees in tumor-bearing tissues of nude mice(P0.05).CONCLUSIONS:DCN can in-hibit the growth of tumor in OSCC nude mi

14、factor receptor;Cellular-myelocytomatosis viraloncogene;Cyclin dependent kinase inhibitorShanghai J Stomatol,2023,32(1):40-46.口腔鳞癌(oral squamous cell carcinoma,OSCC)是来源于上皮组织的肿瘤,也是口腔颌面部最常见的恶性肿瘤,占口腔恶性肿瘤的80%以上1。OSCC早期通常无明显症状,易被忽略,部分患者会出现红色、白色或红白相间的病损。临床上OSCC多呈现为菜花状或疣状外观,也可形成深、大溃疡,癌症晚期可出现疼痛、口腔异味、开口受限、吞咽
15、咀嚼困难、出血和体重下降等症状。目前,OSCC早期以手术治疗为主,癌症后期,单纯手术治疗效果不佳,需联合放、化疗,且某些特殊部位(如舌后部、磨牙后区、口底和软腭)因淋巴结转移率较高,术后5年生存率较低,约为20%40%2。虽然近年来OSCC的治疗方法不断完善,但由于其早期症状不明显,不易做到早发现、早诊断,且由于OSCC在早期易发生淋巴结转移,故易复发,所以对OSCC的研究需追本溯源,明确其发病及调节机制。饰胶蛋白聚糖(decorin,DCN)又称核心蛋白聚糖,因其修饰胶原纤维的特性而得名。DCN是细胞外基质的重要组成部分,是富含亮氨酸小分子蛋白多糖(small leucine-rich pr
16、oteoglycans,SLRP)家族中最具特征性的家族成员。研究3表明,DCN有抗纤维化、抗癌、抗炎和调节血管生成等作用。目前以DCN为靶点的靶向药物治疗已成为癌症治疗研究领域的一大热点,已证实DCN可直接或间接调控癌细胞增殖、凋亡、侵袭、转移和血管生成等过程来发挥抑癌作用3-5。表皮生长因子受体(epidermal growth factor re-ceptor,EGFR)属于型受体酪氨酸激酶(receptorstyrosine kinase,RTKs)家族,是一种跨膜糖蛋白6。研究7-9表明,在多种肿瘤中均可发现EGFR表达升高现象。具体调节机制为EGFR与配体结合,激活下游PI3K/AKT、Ras/Raf、JAK2/STAT3等信号通路,最终调节肿瘤细胞的增殖、侵袭及凋亡等过程10-12。研究13-14提示,在癌症的早期诊断、肿瘤恶性程度判定、术后复发及预后评估方面,EGFR表达水平的检测有一定的指导意义。骨 髓 细 胞 瘤 病 毒 癌 基 因(cellular-myelocy-tomatosis viral oncogene,C-Myc)是原癌基因Myc家族中的一员。研究15
