5、市太和医院(湖北医药学院附属医院)中西医结合科,湖北 十堰442000;2 十堰市人民医院 中医科,湖北 十堰442000 摘要目的:探讨 CircPTTG1IP 调控 mi-223-3p/SMAD3 轴对类风湿关节炎(A)成纤维样滑膜(FLS)细胞增殖、迁移和炎症反应的影响。方法:实时荧光定量聚合酶链反应(qT-PC)、蛋白质印迹法(Western blot)检测正常 FLS 细胞和 A-FLS 细胞中 CircPTTG1IP、mi-223-3p 表达以及 SMAD3 蛋白表达;双荧光素酶实验验证 Circ-PTTG1IP 与 mi-223-3p、mi-223-3p 与 SMAD3 的关系。
6、将 A-FLS 细胞分为 A-FLS 组、si-NC组、si-PTTG1IP 组、si-PTTG1IP+anti-NC 组、si-PTTG1IP+anti-mi-223-3p 组,qT-PC 检测 A-FLS 中Circ-PTTG1IP、mi-223-3p 表达;CCK8 检测 A-FLS 增殖情况;流式细胞术检测 A-FLS 凋亡率;Transwell实验检测 A-FLS 细胞迁移及侵袭情况;Western blot 检测 SMAD3、EMT 相关蛋白表达;ELISA 法检测 IL-2、IL-6 水平。结果:与 FLS 组比较,A-FLS 组 CircPTTG1IP、SMAD3 上调,mi-
7、223-3p 下调;与 A-FLS 组、si-NC 组比较,si-PTTG1IP 组 OD 值,A-FLS 细胞迁移、侵袭数量,Circ-PTTG1IP、SMAD3、N-cadherin、Vimentin蛋白表达,IL-6、MMP-2 水平均显著降低(P 0 05),mi-223-3p 表达、细胞凋亡率、E-cadherin 蛋白表达以454及 IL-2 水平均显著升高(P 0 05);mi-223-3p 沉默减轻了上述指标的变化;Circ-PTTG1IP 靶向调控 mi-223-3p/SMAD3 轴。结论:沉默 Circ-PTTG1IP 可能通过上调 mi-223-3p 来抑制 SMAD3
