2、组织、癌旁正常组织进行研究。分别检测 USP18、ISG15 mRNA及其蛋白表达情况;分析乳腺癌组织USP18、ISG15表达水平与病人临床病理特征、预后的关系;Cox回归分析乳腺癌病人预后的影响因素。结果 Ualcan数据库中乳腺癌组织USP18为18.404.17、ISG15 mRNA表达水平为168.5643.95高于正常乳腺组织(10.003.14、30.768.23)(P0.05);乳腺癌组织USP18、ISG15 mRNA表达水平高低与乳腺癌预后无明显相关性。乳腺癌组织 USP18为 1.670.53、ISG15 mRNA 为 1.860.61及蛋白阳性表达率均高于癌旁正常组织(
3、1.030.34、0.990.33)(P0.05);乳腺癌组织USP18、ISG15表达水平均与淋巴结转移、乳腺癌分子亚型、肿瘤分化程度、TNM分期相关(P0.05);乳腺癌病人癌组织中USP18表达水平与ISG15呈正相关(P0.05);USP18阳性组、ISG15阳性组乳腺癌病人术后60个月生存率均低于USP18阴性组、ISG15阴性组(P0.05);淋巴结转移、TNM分期、USP18、ISG15均是影响乳腺癌病人不良预后的独立危险因素(P0.05)。结论 乳腺癌病人癌组织 USP18、ISG15 表达水平均较高,两者均与乳腺癌病人预后关系密切,检测乳腺癌组织USP18、ISG15水平有助

10、olecular subtype,tumor differentiation,and TNM staging(P0.05);there was a positive correlation between the expression level of USP18 and ISG15 in cancer tissues of breast cancer patients(P0.05);the 60-month survival rates of breast cancer patients in USP18 positive group and ISG15 positive group wer
11、e significantly lower than those in USP18 negative group and ISG15 negative group(P0.05);lymph node metastasis,TNM staging,USP18,ISG15 were independent risk factors for poor prognosis of breast cancer patients(P0.05).Conclusion The expression levels of USP18 and ISG15 in breast cancer tissues of bre

14、5(interferon stimulated gene 15,ISG15)在乳腺癌组织中表达上调,其可能是诊治乳腺癌的潜在靶标3;且USP18可将ISG15从其靶向蛋白上水解出来,进而在调控细胞多种信号途径中发挥作用4。但USP18在乳腺癌病人癌组织中的表达水平与ISG15的相关性及与乳腺癌预后的关系尚未明确。基于此,本研究通过Ualcan数据库检索USP18、ISG15在乳腺癌中的表达情况及二者与乳腺癌预后的关系,并利用实时荧光定量PCR(qRT-PCR)法、免疫组化法检测乳腺癌病人癌组织 USP18、ISG15 表达水平,旨在分析 USP18、ISG15相关性及两者与预后的相关性。1资料
15、与方法1.1一般资料选取2012年5月至2015年7月于渭南市中心医院行根治性切除术的乳腺癌病人99例为研究对象,均为女性,年龄(49.2810.22)岁,范围为3565岁,50岁者53例,50岁者46例;肿瘤长径3 cm的癌旁正常组织),液氮罐中冻存24 h,移至80 冰箱中冻存;剩余部分组织标本放置于福尔马林中固定,用于制备石蜡标本切片。1.2.3 qRT-PCR 法 检 测 组 织 标 本 USP18、ISG15 mRNA表达水平取出“1.2.1”中冻存的标本,研碎,利用TRIzol试剂盒(无锡百泰克生物技术有限公司)提取标本总RNA,应用Revert Aid First Strand
16、cDNA Synthesis Kit(K1622,上海恒斐生物科技有限公司)将组织标本RNA反转录成cDNA。依照Perfectstart SYBR Green qPCR master mix试剂盒(TQ2300-02,北京索莱宝科技有限公司)说明书配制反应体系,运用qRT-PCR仪(ABI7500,上海普迪生物技术有限公司)进行扩增、检测。以 2Ct法计算 USP18、ISG15 mRNA 相对表达量。USP18、ISG15均以 GAPDH 为内参,引物序列详见表1。1.2.4免疫组化法检测组织标本USP18、ISG15蛋白表达水平取石蜡标本切片(厚约4 m),利用链霉亲和素-生物素复合物法染色,参考相关试剂盒说明书进行操作,一抗分别为兔抗人USP18多克隆抗体(上海群己生物科技有限公司,批号 H00011274-B01,稀释比为1 100)、ISG15多克隆抗体(艾美捷科技有限公司,批号A71120-100,稀释比为1 100),应用二氨基联苯胺显色、苏木素复染、梯度乙醇脱水,封片,显微镜(DMi1,上海徕卡显微系统贸易有限公司)下拍照。按染色程度(A)和阳性细胞比例(B)评分乘积
